What is Gram Staining?

The technique was developed by the Danish scientist Hans Christian Gram in 1884 and is still widely used today. Gram-positive bacteria have a thick layer of peptidoglycan in their cell wall, while Gram-negative bacteria have a thinner layer of peptidoglycan surrounded by an outer membrane.

The Gram staining process involves several steps. First, the bacteria are stained with a crystal violet dye. The dye is then washed off and a mordant, usually iodine, is added. This causes the crystal violet to become trapped in the peptidoglycan layer of the cell wall. A decolorizing agent, such as alcohol or acetone, is then added to remove the crystal violet from the Gram-negative bacteria. Finally, a counterstain, such as safranin, is added to give the Gram-negative bacteria a pink color. The Gram-positive bacteria, which still have the crystal violet in their cell walls, remain purple.
